Design of Experiment (DOE) is one of the crucial steps in the setup of a high throughput laboratory. However even people with a strong technical biological or analytical chemistry background may have never heard about it, because it is not taught in all universities. Non-optimal trial and error experimental settings are still ruling many labs.

This very compact tutorial gives an overview about how experiments with multiple input variables (flow rate, buffer concentration, spray voltage, eluent flow) can be planned with a design matrix; are performed in a setup with less experimental runs; are evaluated in a short time with an optimization algorithm.
